Description

In human embryonic kidney cells overexpression of TPP2 induces accelerated growth and resistance to apoptosis, correlated with the appearance of multiple chromosomal aberrations, numerical and structural centrosome abnormalities, and multipolar cell divisions, suggesting a role in the regulation of centrosome homeostasis and mitotic fidelity [2185].

Knockout in mice resulted in viable animals which are almost indistinguishable from their wild type littermates when young. After the age of 1 year mutant mice show an increased mortality and multiple signs of premature ageing. They exhibt an aged appearance, disheveled fur, large bald areas, reduced body mass, and loss of vigor. Mice also show symptoms characteristic of immunohematopoietic senescence, including accelerated thymic involution, lymphopenia, impaired proliferative T cell responses, extramedullary hematopoiesis, and inflammation. Premature cellular senescence in fibroblasts, coinciding with the upregulation of TP53 and the dysregulation of NFKB1 was also observed [2184].
