Observations
The retrograde response is activated in response to mitochondrial dysfunction and extends replicative lifespan. Rho- yeast cells have petite mitochondria and cannot support normal oxidative phosphorylation. Deletion of PHO84 in rho- cells completely abrogated replicative lifespan extension. Overexpression of PHO84 in rho+ cells increased mean replicative lifespan by 14%. In rho- cells mean replicative lifespan was increased by 37%.
